    Partial Replicate Study A partial replicate study is a BE study design frequently used for drugs with high variability in pharmacokinetics (PK). It provides robust insights into intra-subject variability while ensuring compliance with regulatory requirements for BE studies. Key Features of a Partial Replicate Study 1. Study Design: Typically involves three periods (e.g., TRR, TRT, RTR), where each subject receives the test formulation (T) once and the reference formulation (R) twice. This design allows for a more accurate assessment of intra-subject variability. 2. Purpose: To evaluate the variability of both test and reference products. To establish BE using scaled bioequivalence criteria, particularly for highly HVDs with intra-subject variability >30%. 3. Regulatory Focus: Scaled bioequivalence uses widened acceptance limits for HVDs, improving the likelihood of demonstrating equivalence without compromising safety or efficacy. 4. Benefits: Provides robust data for variability assessment. Reduces subject numbers compared to fully replicated designs. Enhances the power to demonstrate bioequivalence for challenging drugs. Data Inference in Partial Replicate Studies: 1. Intra-Subject Variability Analysis: The design captures intra-subject variability for both T and R formulations, which is critical for understanding the differences in individual responses. 2.     Researchers often get stuck in the methods section. They overthink every detail, aiming for perfection. However, here’s the reality: Your methods aren't meant to be a masterpiece of writing. They're a template for others to replicate your study. I've reviewed hundreds of papers. The best methods sections? They're clear, concise, and replicable. Here's how to pull it off with yours: 1. Restate your research question ↳ Remind readers why you're doing this 2. Explain your chosen method ↳ Qualitative? Quantitative? Mixed? 3. Justify uncommon methods ↳ Used something new? Defend it. 4. Detail your data collection ↳ Surveys? Interviews? Be specific. 5. Describe your data analysis ↳ What tools or tests did you use? 6. Justify your methods ↳ Why these specific strategies? 7. Address challenges ↳ What problems did you face? How did you solve them? 8. Consider ethics ↳ How did you protect participants? Your one goal here is to help others replicate your study.   • New chapter on replication studies in my free textbook https://lnkd.in/eB5B9udq  I discuss the difference between direct and conceptual replications, how to analyze them, why don't just do 1 study with alpha 0.0025, and how to deal with conflicting results across studies. I define a direct replication as a study where a researcher has the goal to not introduce variability in the effect size compared to the original study, while in a conceptual replication variability is introduced intentionally with the goal to test the generalizability. I hope this is helpful. Some approaches focus on a judgment of the similarity in operationalizations between studies, but as there are always differences, I think we should focus on what the researcher intends to test.   Then I discuss 3 approaches to analyze replication studies. A test of differences in effect sizes. Is the replication study significant? Is the effect (or the difference between effects) too small to matter? The first is too rarely done, but the most interesting. Then it becomes a bit niche, but I dive into why we do 2 studies with alpha = 0.05, and not just one study with alpha 0.05 * 0.05 = 0.0025. There is a bunch of non-statistical reasons (identifying systematic error).   Interestingly, it depends which of the 2 options is more efficient, in terms of that you need a smaller sample size. It does not matter a lot, but depending on the power, sidedness of the test, and effect size, either can be just a bit more efficient.   Then I discuss Many Labs 5 as an example of how difficult it is to predict if studies will replicate. It’s a great study showing researchers can’t predict whether moderators matter in replication studies. It shows we can only know is something replicates if we replicate it.     Replication is not optional, it's foundational. Here I go again with another reason I often reject manuscripts without review as EIC. In plant pathology (and most fields), repeating experiments over time should be standard and planned in advance. Some authors overlook a basic requirement in our Instructions for the Authors: inform that the experiment was replicated at least once (eg. different times or locations) and how all data was incorporated into the analysis. It’s not enough to say an experiment was repeated; results must be analyzed as a whole. Use appropriate tools: include "experiment" as a factor and test for interactions, assess variance homogeneity, or apply mixed models or meta-analytic approaches. Selecting the “best” or most expected run undermines scientific rigor. Cost constraints are real, but not an excuse for poor design. Use efficient designs, pilot studies, power analysis, historical data, or collaboration to optimize resources. If full replication isn't feasible, justify and discuss limitations - transparency is always better than omission!
