How do you handle changes and deviations during and after the validation process?

During and after the validation process, handling changes and deviations becomes even more critical. During validation, any changes should be carefully assessed to ensure they do not compromise the ongoing validation efforts. It is to make sure we are not fixing one thing and accidentally breaking another in the process. After validation, the focus shifts to maintaining the validated state. Dealing with deviations involves investigating and addressing any unexpected deviations from the expected performance or compliance. Overall, a robust change control process ensures that whether we are in the middle of validation or maintaining a validated system, changes are managed in a controlled and documented way.

In my experience, Change control in GxP ensures safe updates to systems. It starts by defining the change, who's in charge, and looking at risks. Testing and documentation are key to make sure changes won't harm the system. Once approved, execute the plan and document results. Reviewing the change is important to confirm it works as intended. Good change control follows rules, involves everyone, and keeps systems reliable and compliant with regulations. Regular training helps teams adapt, making sure the process stays effective.

When changes or deviations pose a high impact during or after validation, they should be locked following established procedures. Additionally, periodic maintenance and verification processes help maintain compliance over time.

Change Control is a process to document any change in a validated system or process in order to document the proof of things which have been changed in a validated process or system and after those changes validated system or process have the validated state maintained.

Quality Risk Management (ICH Q10) should be utilised to evaluate proposed changes. Change control should have risk assessments performed by the affecting departments (e.g. Manufacturing or Bioanalytical or both) including Validation and QA. Risk assessments should highlight the impact on the product quality and patient safety. A clear objective and timeline should be defined in the change control.

In my view deviation is an opportunity to improve the quality system and action derived from RCA will help to avoid future occurrence.

Deviation Management helps to handle if something unexpected happens, how to deal with it quickly. Both work together, making sure all changes, whether planned or unexpected, are taken care of properly.

Deviation Management is a process of identifying, managing via remediation, correction and preventive of unexpected conditions to avoid potential noncompliance and/or noncompliance in a GMP environment.

Effective deviation management ensure companies are in compliance with the regulatory standard and local policies. This is a part of continuous improvement and companies can improve their procedures, training methods, etc via effective deviation management. Identifying a true root cause can help company improves the current procedures. It's easy to identify human / man as a root cause but a root cause analysis should not stop there. If man / human is identified as a cause for RCA, investigation should research deeper about procedures or training.

In my view, handling deviations in GxP systems is important. Investigate thoroughly, find the main issues, and fix them appropriately. Make sure the solutions match how severe the problem is. Keep a close eye on things, document well, and follow a strong process to close the case. This way, we ensure the system works reliably and stays in line with the rules.

First and foremost, every organization has their own individual procedures on Deviation management. It’s NOT one-size fits all approach! Deviation during the validation needs to be addressed/resolved and documented within that validation deliverables like Summary reports, Trace Matrix and Final Report. Whereas, deviation after Validation can be handled using deviation management procedures.. Risk analysis might help to determine the level of documentation afterwards. It can be a CAPA or simple testing with revised traceability of the protocol or it can be an Event that needs thorough investigation.

Keeping a system validated is important, even after the first validation. The time when the system is actively running, called the operational phase, is usually the longest. To make sure everything stays in check, we use controls like release management, handling incidents, checking things periodically, and getting support from vendors. Following the rules about protecting data, keeping good records, and checking things regularly are super important. It's like giving the system regular check-ups to make sure it's working well.

Some recommended best practices to handle the changes are: a) Have a well-defined change control process b) Keep clear change request documentation c) Conduct risk assessments d) Perform testing and validation of changes e) Maintain comprehensive documentation and record-keeping
